Study of some processing changes in proteins
Requirements (Instruments, Chemicals & Other) :
- Protein powder
- Analytical Balance
- Conical flask
- Petri-dish
- Whatman filter paper
- Convective hot air oven
- Shaker
- Buffer solution
- Measuring cylinder
- Distilled water
Procedure :
- Weigh 10g protein powder (S) in five different conical flasks.
- Pour 100 ml buffer solutions of pH 3.0, 4.0, 4.5, 6 and 8 in the five different proteins containing conical flask and marked as the A, B, C, D and E respectively.
- Shake the flasks at 200 rpm for 10 min.
- Filter the solution of each flask using Whatman filter paper.
- Transfer filter paper containing insoluble matter inside the pre-weight petridish.
- Put the Petridish in the oven and dry the content at 105°C for 3 hrs.
- Record the weight of petridish after 3 hrs of drying.
- Weigh Whatman filter paper and record the weight.
- Calculate using formula:
- Draw curve solubility (%) Vs pH.
